• Somatic Embryogenesis Definition : “Somatic embryogenesis is an advanced micro-propagation technique in which a plant or embryo is derived from a single somatic cell or group of somatic cells. Somatic embryos are formed from plant cells those are not normally involved in the development of embryos. ”
• Advantages of Somatic Embryogenesis : Ø Clonal propagation of genetically uniform plant material. Ø Elimination of viruses. Ø provision of source tissue for genetic transformation. Ø Generation of whole plant from single cell. Ø Development of Synthetic seed technology.
We do Somatic Embryogenesis specially for Douglas Fir; one of the tree species having very high commercial importance- Ø Douglas Fir is used as a plantation tree for timber in Europe, North America and many other countries. Ø It is extensively used in construction of Air crafts, ships and houses due to light weight and high strength of wood. Ø It is also used in Paper industry for extracting Cellulose fibres. Ø It is also used as Christmas tree.
The steps involved : - Ø Initiation Ø Embryonal suspensor mass cultures and multiplication Ø Cell Suspension culture Ø Embryonal cells Cryo-preservation Ø Embryo Development Ø Embryo packing and shipping Ø Embryo plucking for the germination and Emblings production ØEmblings harvesting, packing and shipping.
Initiation : ØWe import the viable seeds with Cones from US every year. ØAfter implementing different treatments, we dissect out the Zygotic embryo from the seeds under highly sterile conditions. ØAfter few weeks of incubation on specific nutrient medium; the proliferation starts and we get the complete mass of Embryonal cells (ESM).
Embryonal Suspensor Mass (ESM) Cultures : - ØMaintenance and multiplication of ESM is being done by transferring ESM on fresh medium (solid) every 10 -12 days. Ø We are maintaining around 10 thousand ESM cultures in our growth rooms. ØAll cultures are being incubated in darkness at 22 -25 O C.
Cell Suspension Culture : ØThe cell suspension culture starts by the addition of ESM cultures in liquid medium. ØContinuous rotation on shaker machine at 80 -100 rpm in dark and sub-cultured at approximately 7 days interval. ØWe maintain n multiply over 2000 flasks of Cell suspension culture at every week.
Cryo-preservation Facility : - Ø we preserve the Clonal Embryonal cells in liquid Nitrogen at minus 196 O C temp. ØWe can preserve the clones over 10 years.
Embryo Development : ØThe cotyledonary embryos are developed after 8 weeks of incubation from Embryonal cells on development medium. ØEmbryos are developed in dark at 25 O C.
Somatic Embryos: packing and shipping Ø Embryos are transferred on Petridishes with nutrient medium and then packed in thermocol boxes with ice-packets for shipping.
Embryo germination and Embling production : - ØEmbryos are germinating in light. Ø after 8 -10 weeks Emblings are being produced and are ready to harvest.
Emblings harvesting, packing and shipping : ØGood Quality emblings are harvested and packed in Petri-plates with sterile water. Ø 50 Emblings are packed in one Petriplate; and then Petriplates are packed in thermocol boxes.
