The essential and mysterious role of Themis in thymocyte development Dec. 14(Wed) 11: 00 / Auditorium (1 F), PBC Guo Fu Xiamen University Themis, thymocyte-expressed molecule involved in selection, is selectively and highly expressed in T cells. Detailed analysis showed that Themis expression is developmentally regulated in thymocytes and inducibly up-regulated in mature T cells upon activation. These expression profiles suggested that Themis may play an important role in both thymocyte development and peripheral T cell function. Using Themis germline knockout mice we showed that Themis controls thymocytes positive selection. Mechanistic study revealed that Themis functions as a negative regulator by dampening TCR signaling amplitude and exerts its function in a narrow TCR stimulation strength. We also investigated the role of Themis in mature T cells. By breeding Themis “floxed” mice to d. Lck-Cre mice, we could specifically deleted Themis expression in mature peripheral T cells. In this scenario we studied the function of Themis in various infection models and other experimental settings, and we found that Themis is both required for T cell homeostasis and T follicular helper cell differentiation.
