- Slides: 19
• Step #1: Always carry with 2 hands
• Step #2: Only use lens paper for cleaning
• Step #3: Do not force knobs
• Step #4: Always store covered
• Step #5: Keep objects clear of desk and cords
Eyepiece Body Tube Revolving Nosepiece Objective Lens Stage Clips Diaphragm Light Arm Stage Coarse Focus Fine Focus Base
COPY! STEP #1: Take a sheet of lens paper and gently wipe each objective lens. STEP #2: Prepare your slide (wet mount) or choose an already prepared slide. STEP #3: Make sure the microscope is on the lowest objective lens (scanning 4 X magnification) STEP #4: Turn the microscope ON. STEP #5: Make sure the diaphragm is open to the maximum setting. STEP #6: Place your slide onto the stage and secure your slide with the stage clips. STEP #7: Move the slide around until you see the specimen while you are looking through the eyepiece. STEP #8: Use the coarse adjustment knob to bring the specimen into general focus. You may change your objective lens to low power (10 X magnification) at this time. STEP #9: Once your specimen is in focus, change the objective lens to high power (40 X magnification). STEP #10: Use the fine adjustment knob to bring the specimen into sharp focus. STEP #11: Sketch the specimen in your notebook or on your lab paper. STEP #12: Remove the slide from the stage STEP #13: Wipe each objective with more clean lens paper. STEP #14: Turn off the microscope STEP #15: Unplug the power cord and wrap it around the base of the microscope. STEP #16: If there is a cover, place it on the microscope. STEP #17: Return the microscope to a clean cabinet
How do you calculate the total magnification of a specimen? COPY! Eyepiece lens has a magnification of 10 X. Objective Lenses ◦ ◦ Scanning Lens has a magnification of 4 X. Low Power lens has a magnification of 10 X. High Power lens has a magnification of 40 X. Oil Immersion lens has a magnification of 100 X. Total Magnification = eyepiece magnification x objective lens magnification

Microscope Field of View COPY! • The diameter of the field of an optical light microscope is the field number, which is the diameter of the field view in mm measured at the intermediate image plane. • The field number is typically listed on the microscope eyepiece. • The higher your magnification, the smaller the microscope field of view will be.
