Sharing the KOMP Resources Mutant Mouse Repositories Clonal ES cell lines NARRC January 2007
History of Mouse Resource Support Ø 1959 - NSF funded 1 st JAX mouse repository Ø 1978 - Division of Research Resources (pre-NCRR) funded the JAX mutant mouse repository Ø 1999 - NCRR funded MMRRC to expand mouse resource capability Ø 2006 - NIH funded • Repatriation of targeted mutation strains • Acquisition of Deltagen/Lexicon strains NARRC January 2007
Value of KOMP Resources Ø Provide mutant mice and ES cells for • Basic research to basic & clinician researchers • Understand pathways • Understand gene function • Identify drug targets • Phase 0 drug trials Ø Critical to archive and distribute knockout mouse resources NARRC January 2007
Size of current NCRR resources JAX 935 MMRRC 41 Total 976 Mouse strains - cryo. 1, 993 1, 013 3, 006 ES cell lines * 14, 273 Mouse strains - live *JAX C 57 BL/6 J ES cell line distributed by ATCC. TOTAL NARRC January 2007 2928 15, 327 18, 255
Partnerships for Sharing Resources Ø FIMRe - Federation of International Mouse Resources • • Coordinate mouse resource archiving and sharing Provide each other local distribution centers Standardize protocols Share advancements and training Ø IMSR - International Mouse Strain Resource • One-stop on-line shop for mouse resources available worldwide NARRC January 2007
Maintenance of resources Ø Living mice are not necessary - impossible except for strains in continuing demand Ø Gametes & ES cells are mice in different formats • 2/3 of strains at JAX in frozen state only • ~96% of strains in MMRRC in frozen state only Ø Both resource programs cryopreserve strains during the importation process Ø C 57 BL/6 sperm can now be reliably frozen and recovered by IVF Ø Frozen sperm enable recovery of large numbers of mice NARRC January 2007
