Microscope Important Lab instrument
Why use a microscope? n n To view objects and detail too small to see with human eye. Improves resolution of object List examples of how what you used it for: ¨ Blood n cell detail rbc, wbc, platelets ¨ Bacteria n Cocci, rod, bacillus ¨ Protozoa n n Trichomonas Giardia
Types of Microscopes n Compound microscope ¨ Bacteria, n Electron microscope ¨ Required n fungi and protozoa for viruses Fluorescence ¨ Used as a diagnostic tool for immunofluorescence tests
Parts of a microscope n Coarse adjustment ¨ 1 st step in focusing to change the distance between specimen and lens n Fine adjustment ¨ To fine tune the picture ¨ Used particularly with 100 x and oil objective n Stage ¨ Holds the slide, moved up and down with coarse and fine adjustment
Parts of a microscope n Objectives ¨ Common objectives are 4 x, 10 x, 40 x, 100 x, oil ¨ Total magnification is eye piece magnification multiplied times objective you are viewing with n 40 x objective and 10 x eye piece is 400 x magnification ¨ Oil objective is labeled and is always used with immersion oil n Oil increases the resolving power by focusing the light rays
Proper care for microscope Always start with lowest objective to focus n Always store with lowest objective locked in place n Carry with two hands n Cover with dust cover n Be sure to clean oil off of oil objective n Use fine adjustment with 100 x and oil objective n
THE COMPONENTS OF THE MICROSCOPE What do you remember?
THE MICROSCOPE n n n EYEPIECE What is it when there is only 1? When there are 2?
THE MICROSCOPE n ARM
THE MICROSCOPE n BASE
THE MICROSCOPE n TUBE
THE MICROSCOPE n REVOLVING NOSEPIECE
THE MICROSCOPE n n OBJECTIVE LENS What are the magnification of the 4 objectives? ¨ 4, 10, 40, 100 x ¨ 10 x unless otherwise stated What magnification is the eye piece? What would be the total magnification when viewing an object through the 40 x objective? ¨ 400 x
THE MICROSCOPE MECHANICAL STAGE n Why is a mechanical stage better than using clips? n
THE MICROSCOPE IRIS DIAPHRAGM n What does this do? n ¨ Control the amount of light
THE MICROSCOPE COARSE ADJUSTMENT KNOB n What 2 objectives do you use the coarse adjustment? n ¨ 4 and 10 x
THE MICROSCOPE n FINE ADJUSTMENT KNOB n n What objectives is this used with? All, but mostly 40, 100 x
THE MICROSCOPE n. Light
THE MICROSCOPE n ON/OFF SWITCH
CARE OF THE MICROSCOPE n Always carry the microscope by the arm and the base and use 2 hands!
CARE OF THE MICROSCOPE n Wrap the cord up or around the microscope so it is not hanging down.
CARE OF THE MICROSCOPE Always lower the stage and clean all objectives off n Always use a dust cover! n
