Mallory's PTAH stain 1) 원리 : Hematoxylin과 Phosphotungstic acid를 혼합하면 Lake를 형성하여 Muscle의 cross Striation에 염색되고 남은 Phosphotungstic acid ion은 collagen fiber에 염색된다. 2) 염색액제조 (1) PTAH(phsphotungstic acid hematoxylin, 20 : 1) Hematoxylin(CI. No. 75290) 1 g + Phosphotungstic acid 20 g + D. W 1000 ml 각각의 염료를 증류수로 용해시키고 혼합하여 3 - 4주 정도 공기 중에 산화시킨 후 사용한다. 즉시 사용해야 할 경우 pot. permanganate 0. 2 gm을 첨가하여 인공 산화시킴 (2) 3% Pot. dichromate : pot. dichromate 3 g + D.
