What is inside the extraction solution Sodium Chloride Na. Cl Detergent Solution In this experiment, sodium chloride acts as an enzyme to cut proteins just like a pair of scissors. As you should know the cell has two membranes made of phospholipids (fat). The detergent attaches the lipid because of their similar structure. The DNA in the nucleus of the cell is molded, folded, and protected by proteins. The sodium chloride cuts And allow the DNA to float in the proteins away from the solution DNA.
Step 1: Separating the cells Put the mashed fruit into a becker Add 10 ml of extraction solution, composed of sodium chloride and detergent solution Mix for 5 minutes Strain the solution to remove thickest part Pour the mixture into a test tube
Step 2: Alcohol Separation Tilt your test tube and slowly pour the ethanol into the tube down the side so that it forms a layer on top of the cell mixture. Pour until you have about the same amount of alcohol in the tube as cell mixture. Alcohol is less dense than water, so it floats. • As you can see the DNA is already visible here
Why did we add alcohol? DNA normally stays dissolved in water, but when salty DNA comes in contact with alcohol it becomes undissolved. This is called precipitation. The physical force of the DNA clumping together as it precipitates pulls more strands along with it as it rises into the alcohol. You can use a wooden stick or a straw to collect the DNA. If you want to save your DNA, you can transfer it to a small container filled with alcohol.
Step 3: Extraction Use the loop of glass to take the DNA out of the test tube. Do it slowly and carefully to not damage the result • Extracted DNA
