Recombination DNA What is recombinant DNA ? ? • Recombinant DNA ; 두 개 이상의 source에서 유래한 DNA가 결합된 artificial strand of DNA. the basis for all sorts of genetic engineering. In vitro recombination 1 1. Insert와 vector를 동일한 Restriction enzyme을 이용하여 자른다. (잘린 부분은 서로 complimentary single stranded sections을 가짐. ) 2 2. Complimentary sticky ends Pair끼리 서로 만나면, 3 3. Restriction Enzyme Restriction enzymes(restriction endonuclease)는 DNA 내 특정 염기서열을 인식하고 절단하여 double-stranded cut을 만든다. restriction enzyme의 종류 1. Type I enzyme ; 3개의 subunit으로 이루어져 있으며 target site를 인지한 후 target에서 조금 떨어진 부분의 non-specific site에서 restriction을 유도. ATP가 필요 하다. 2. Type II enzyme ; target site를 인지하면 그 site 내부를 자르게 된다. 대부분 palindromic sequence를 인식. sticky end 또는 blunt end를 유도. ATP는 필요없다. sticky end Blunt end 3. Type III enzyme; 2개의 subunit으로 이루어져 있으며, target site를 인지한 후 recognition site로부터 24~26 bp downstream에 떨어져 있는 site를 자른다. 6

2. 2 DNA elution from agarose gel ü electrophoresis한 gel에서 원하는 부분의 DNA band를 얻는 방법. ü Et. Br로 염색한 DNA를 UV상에서 보고 자르는 작업이므로 빠른 시간 내에 해야 UV로 인한 DNA damage 를 최소화할 수 있다. ü주의: UV light, Et. Br DNA elution Methods 1. DEAE-Cellulose membrane 이용 agarose gel 안의 DNA를 membrane에 붙게 한 후 elution 2. electroelution dialysis bag 에 gel을 넣어 electrophoresis한 뒤 Agarose로부터 빠져나온 DNA 농축 3. Gene Clean Kit 이용 DNA와 결합하는 glassmilk (silica matrix)를 이용하여 DNA를 신속하게 분리 18

Protocol of DNA purification Digested Vector 1. Add 500µl of BNL buffer to Enzyme mixture and Mix well by pipetting Binding of DNA 1. 2. 3. 4. mini column을 collection tube에 insertion한다. Column을 new 1. 5 ml tube로 옮기고 50 µl of Distilled water RT, 1 min incubation → 13, 000 rpm, 1 min 19

Protocol of DNA elution from agarose gel Gel slice (DNA 함유하고있는 gel) 1. Agarose에서 원하는 밴드를 잘라 1. 5 ml tube에 넣는다. 2. Add 100 µl of membrane binding buffer per 100 mg of gel slice (우리의 gel 의 무게는 200 -300 mg 가량 되므로 600 ul의 buffer를 넣는다) 3. Vortex & Incubate at 50 -65 ℃ (gel이 완벽히 녹을때까지) Binding of DNA 1. 2. 3. 4. mini column을 collection tube에 insertion한다. Dissolved gel mixture를 column으로 옮겨준다. 1 min, RT, incubation 13, 000 rpm, 1 min → Flowthrough 제거하고 column은 다시 제자리 Washing 1. 2. 700 µl of membrane Washing buffer → 13, 000 rpm, 1 min → Flowthrough 제거 Column only → 13, 000 rpm, 1 min Elution 1. 2. 3. Ligation 두 DNA strand의 sticky end에 hydrogen bond (A-T and G-C pairing)가 형성되면 LIGASE가 energy source를 이용하여 phosphodiester bond를 연결시켜서 DNA를 결합시킨다. 21
